Sprouting and Dedifferentiation of Excised Gladiolus Cormel Tip in Vitro


Abstract
This study was carried out to investigate the relationships among the change of endogenous substances, sprouting and dedifferentiation of gladiolus cormel during the storage. ABA-like substances existed in gladiolus cormel so much after harvest but, as time goes by, ABA-like substance contents were decreased and GA-like substances were appeared 110 days after cormel havest. Starch and sugar contents by the storage temperature, starch existed so much in gladiolus cormel than others and the next was non-reducing sugar. Reducing sugar existed a little. Non-reducing sugar contents was increased a little by the storage time went by but starch and reducing sugar contents changed little. 100 days after cormel harvest, requiring days for cormel tip sprouting was shortened from to 4 days. Days required for sprouting of Topaz was 7, Firebrand, Hunting Song, Nova Lux and Red Beauty was 10, and Violet was 1. days. Potential of dedifferentiation of cormel tip was remarkably increased 140 days after cormel harvest and 10¡É (night)¡ê20¡É (day) treatment was more effective than 5¡É treatment for callus induction. Sprouting of cormel tip was inhibited by the existence of bracts and inhibited remarkably in the dark.
